マップチップのドットの向きを反転させたりしたいのですが、向きは固定なのでしょうか？

エディタ上の操作ではそれは出来ないと思います。（向きは固定ということです）

RPG MAKER UNITEと関係のないグラフィックツールなどを使って、例えば左右反転したマップチップの画像を作って保存すれば、RPG MAKER UNITEで読み込むことができ、変更されたマップチップを使えると思います。
